To identify the presence of sperm in the vaginal cavity, forensic scientists typically use a combination of tests and procedures. These include: 1. **Microscopic Examination**: Direct observation of the sample under a microscope to detect the presence of sperm cells. 2. **Acid Phosphatase Test**: A biochemical test that detects the enzyme acid phosphatase, which is present in high concentrations in seminal fluid. 3. **Prostate-Specific Antigen (PSA) Test**: Also known as the p30 test, this test detects PSA, a protein produced by the prostate gland and found in seminal fluid. 4. **DNA Analysis**: Forensic DNA profiling can confirm the presence of male DNA, which can be further matched to a specific individual. The acid phosphatase test and the PSA test are particularly useful in confirming the presence of seminal fluid even if sperm cells are not found or are degraded.
