Can a defamation case be filed is someone spreads false rumors

Suggestion needed.. Including my wife they are 4 siblings(sister 1,sister 2,my wife and brother).. Sister 1 got pregnant with test tube baby in 2014 after sister 2 husband has given his seed and has love relations with sister 2 husband, Sister 2 is not aware about that and my wife has hint but she didn't told any body.. Last year my wife told this to sister 2 about this and there was a big argument and later on sister 2 got clue that her husband has relations with sister 1 and now Sister 1 husband is also saying my wife is wrong and he is threatening us that he will file a case against my wife and will defame us.. Can he file a defame case?

Sorry but I found your explanation confusing and clarity is required to give proper advise. But generally speaking defamation can be filed as Civil or Criminal case and evidence is required for it. It is very difficult to prove spreading of verbal false rumours.
