If your father is alive, then no need, it is sufficient if your father signs.

If your father died before 2005, and you have no brothers, and your mother is alive, then both you and your mother must sign.

If your father died before 2005, and you have no brothers, and your mother died after 2005, then your sister must sign.

If your father died before 2005, and you have no brothers, and your mother also died before 2005, then it is enough if you sign.

If your father died after 2005, and your mother is also dead, then your sister must also sign. If your mother is also alive, then you, your sister, and your mother must sign.

Your father's brother have no share in the property legally.

Sir/Madam,

If your father planning to sale his self acquired property, you and your no need to sign, provided if the buyer wants that you and your sister has to sign, in the said property your father's brother cannot claim any share, since it is self acquired.

Your question is not clear. Better write in your own language so that you will have the comfort. However in respect of self earned property any male person under Hindu Law can do anything with the said property.
