Can my cousins claim part in agricultural land exclusively in my single name for last forty years? They are doing cultivation on my land at present but now I want my land back. They say they will file suit against me court as they are cultivating the land for last many years.
No, the land is on your name and you are the owner. Mere irrigating does not entitle them for getting land transferred on their name.
It is suggested to personally discuss with them to vacate the land after the current crop. Else you can take over the possession yourself after the current crop.
Or else if you want you can sell and move out of this land

Yadi vo jameen sirf aapke naam se hai aur koyi bhee us per kabja kiya hai to aap tatkaal pahle apne documents ke saath police ki sahayta le. Yadi baat nahi banti hai to civil court me bedakhli ka mukadma dakhil kare.
You are the owner of the land. Cultivation by cousins, if any, was on your behalf. They were only licencee. Therefore, mere cultivation will not make them owners. But will have to see whether it falls within exceptions to provision which prohibits letting by tenure-holer under UP Z.A. & L.R. ACT.
